Yes - this is perfectly possible.

Do you have an SMTP Connector or just an SMTP Virtual Server?

If you are using the SMTP Virtual Server only, right-click it and choose properties, then on the Delivery Tab, Click on the Outbound Security Button and fill in the relevant credentials.

Staying on the Delivery Tab, click on the Advanced button and fill in the Smarthost field.

Restart your Simple Mail Transfer Protocol Service and test outbound mail-flow.

If you are using your SMTP Connector, right-click and choose properties, then on the Advanced Tab and then on the Outbound Security button and fill in the relevant credentials.

Then on the General Tab, fill in the "Forward all mail through this connector to the following smart host" field with the relevant Smarthost details (FQDN or IP Address wrapped in Square Brackets e.g., [123.123.123.123]).

Again - restart the Simple Mail Transfer Protocol Service and test outbound mail-flow.
