After your vm has been powered off you will right click on the vm and hit migrate. Next you will pick change datastore. Last you pick the datastore to send it to. That is it.

Have good backup. I am not the person to answer that but when I work for a hosting company. I did 100's of moves and never ones had an error.

Do you have a cheap nfs or iscsi NAS that you can clone to. That would give you a backup.

During off hours make a clone to NAS then move your VM to the new datastore. If s*it hits the fan you can use the clone to restore from.

Before moving why not just create an 8MB blocksize for the datastore, it does not cause any performance issues.

Turn OFF VM, and follow my EE Article.

HOW TO: Clone or Copy a virtual machine in VMware vSphere Hypervisor ESX/ESXi 4.x or ESXi 5.0

or Right Click the VM, and Select Migrate.
