pupbld

I cannot run the script

SQL> @/u01/app/oracle/product/11.2.0/db_1/rdbms/admin/pupbld.sql
SP2-0310: unable to open file "/u01/app/oracle/product/11.2.0/db_1/rdbms/admin/pupbld.sql"
SQL>

Is there any other options..

Try:
@/u01/app/oracle/product/11.2.0/db_1/sqlplus/admin/pupbld.sql

Also, there is a sqlplus shortcut to ORACLE_HOME:

@?/sqlplus/admin/pupbld.sql

Actually why we are running pupbld.sql

Were you connected as SYSTEM?

If not, then you should have been.  If you look in Oracle provided scripts, they will normally tell you what you need to do.

In this script it has:
--   Connect as SYSTEM before running this script

I give up, why?  You must have been told to run that script by something or someone.

If you are asking why run this, the script builds the default users for the database.

Not true.

If you open that file and read it, it tells you what it does:

--   Script to install the SQL*Plus PRODUCT_USER_PROFILE tables.
