VB.net using Windows API

Ran across this comment below from another sight, and

Having trouble trying to grasp the concept of how to do this with VB.net
"Grab an instance of the object in your code, then control the object. With VB and Internet explorer, you can do almost anything to any webpage. I use this a lot at work to pull a ton of reports for a web based system."

How do you 'grab' an instance of an object with the code?
Then how do you proceed with controlling the object?

Having trouble understanding Windows API and how to use it with VB.net
Can anyone shed some light on this? or give advice on it, so I can fully understand it.
