Is there any vulnerabilities for Server and Domain Isolation

I heard mostly positive of applying Domain isolation as a good security measure. Is there any vulnerabilities at all?

I have a site that have 100s of buildings,  they are all connected into a common network.  Physically they are not in one single fiber but rather in network segments with router and switches to make them into a single network.
Now I am evaluating a possibility of creating a more secure separate network without wired another fiber across all the buildings.  The network will consists of a computer at each building.   I am thinking a Domain Isolation and like to understand the drawback for someone that have experience in this area.  Or is it better to modify the switch/router AC:L configuration to create a VLAN.  Will such a VLAN secure?

So if we can lock down the USB drive and other removable media and as well as installing uptodate anti-virus scan.  The Isolated network will be as strong as steel if no one from inside sabotage the system.
