You need to use the following ESC codes

ESC p m t1 t2

Outputs pulse to Drawer kick-out port.

m = 0, "0":connector pin 2
m = 1, "1":connector pin 5
t1: on time (* 2ms)
t2: off time (* 2ms)

from this link: http://download.epson-biz.com/modules/pos/index.php?page=single_doc&cid=3087&dcat=27&pcat=3

sample codes can be found at this link: http://utilitysoft.com/CashDrawerCodes.pdf

Then you can see an example of VB.Net to USB with Canon POS printers at this link: http://www.mindfiresolutions.com/How-to-Print-Receipt-by-POS-Printer-using-VBNet-782.php

OR , if you have not done so, download the EPSON OPOS ADK for .NET  from this link: http://download.epson-biz.com/modules/pos/index.php?page=soft&scat=39  This is the best option as it allows you to do all sorts of these things with ease using VB.Net.
