Use the following Backup :
stsadm -o backup -url http://sharepoint -filename d:\backup\Backup.dat -backupmethod Full -overwrite

It doesn't return any error. Afterwards, I use the following command to restore:
stsadm -o restore -url http://sharepoint -filename d:\backup\Backup.dat -overwrite

It fails with the following error message, currently my whole sharepoint server can't access. Any idea ?

This is the problem with permission from where you are taking backup and where you are restoring.

Check that hope that helps

Currently the sharepoint cannot be connected. Sharepoint 80 is started. SQL and Sharepoint services have been started.

How to troubleshoot which part of SharePoint cause the problem ?

Download ULS Log Viewer then open your current ULS Log file with the help ULS Log Viewer than try to open SharePoint with keep opening ULS Log viewer you will find some  correlation ID on your SharePoint with that correlation ID find in ULS Log View and see what exact error it is.

If you can't understand than just over here we will help on it.

Are u talking about the setting under Central Administration  Diagnostic Logging  ?

I have clicked SharePoint FOundation and set Least critcal event & Least critcal event set to verbose.

Open a Sharepoint and the LogViewer. Nothing come up.

Is the viewer only open a log created by SharePoint, or it will online monitor any event (currently nothing there) ?
