SharePoint 3 to Sql Server 2008 R2 application pool error

About a year ago, we rolled out SharePoint 3.0, which connects to a SQL Server 2008 R2 database.  We created a generic account to use in order for the Sharepoint application pool to connect.  It had been working up until yesterday morning at about 0300, and now I keep getting a TCP error, which kills the sharepoint application pool in IIS - it generates an HTTP 503 error when trying to open the page in a browser.

After some experimenting, I discovered that SharePoint was able to establish a connection with my domain admin account.  The generic account is in Domain Users (W2K8 R2 domain) and IIS_WPG.  Any ideas on why this stopped working all of a sudden?

No changes have been made to either the web server or the database server, and the only thing the DB server is used for is the back end for SharePoint.  The web server only hosts SharePoint.
