I'm using Exchange Server 2010. What is the best method/software to backup my mail stores?

What are the steps it takes to use Symantec Backup Exec to backup Exchange mail stores? Do the mail stores need to be unmounted before backup?

No, You do not need to unmount mailbox using Symantec backup exec. Moreover in this Full backup it will also flush logs that are useful for space on disk.

If the Exchange server is on Vmware virtual environment then you can use Veeam backup for taking backup of full server. It includes the functionality of Exchange explorer which very cool for restoration process.
