Group Policy Inheritance Question

If I block inheritance of all domain GPOs to an OU, can I then link a desired GPO to the OU and have it function?

Yes you can, but if there are any enforced policies in the parent OUs those will still flow down. You can check those by selecting OU and Group Policy Inheritance Tab from GPMC.
