Lync Desktop Client

We are looking to start using Lync Online.

Other than the Lync software which is installed on the workstations, is there anything else that's required to use the Lync system, e.g. a minimum version of office (we are using Office 2010). Do we need to install the desktop setup tool software?

you need the Sign In Assistant, which is installed when you run the desktop setup. Or you can manually deploy it:

Is it mandatory or a nice to have? I can see from the link that it helps with connectivity so the users don't have to use their username and password, but does it have to be deployed for it to work?

It will work without it, but not really recommended. Mandatory if you have AD FS.
