WSS_Content

Can we attach one more WSS_Content database to an existing web application, eg http://portaldev/ ? If yes, what's the normal application for this ?

If there are two database attached to it, does the web application use either one or both of them.

Central Admin -> Application Management -> View All Site Collections

The Content DB for each Site Collection is listed.

You can have multiple content databases, but only one per site collection. (Many per application)

Relationship is one web application can host only one content database.  To add to this, you cannot make a copy of a content database and then attach it to a different web application on the same server (some people want to have a test environment on the same server)

It's possible to have 500 content DBs per Web Application;

You can have 500 content database per web application. But your site collection will use only 1 database. 1 database per site collection. In your case , http://portaldev/ is your root site collection which already has a database.Adding one more will not help.
In case you are creating one more site collection in that web app then you can go and create new database from central admin > operations > manage content database > Add a database. Give a name and it would get created in SQL. Bring Wss_content in offline mode.Create a site collection and then you would find new site collection will use new database. Now you can bring WSS_Content in ready mode.

There are two content database mounted to http://portal:8888. How can I know which content database does "8888" use ? Tks
