All suddenly I cannot access my intranet site and my users are complaining too.. What are some of the basic steps I should take to resolve issue?

Run command prompt as administrator
Type  iisreset

Also you could check an application pool in IIS; or services running; or check your database server connection;

Do check event logs for any error messages
