We are running exchange 2010 and want to know if there is a way to create a retention policy based on file size emailed instead of days. We have users that mass email large files and we want to purge those emails only.

Not possible to select on size for retention policy's.

you better limit this on transport level (organization -> hub transport -> global settings). This way they can not send extreme large files.
