In a corporate network employees do not have access to the internet.
They are setting up an Intranet on a local server on IIS.
If there is a youtube video embedded on the intranet can it be accessed by the employees.

If the YouTube video resides on YouTube servers then they reside on the Internet and your users will not have access to it.  The only thing they will be able to access are what is on the Intranet server.  If you need content available to users from the Internet then I suggest you deploy a proxy solution where you only allow access to YouTube.
