Will Autodiscover reconfigure an existing Outlook profile?

We are migrating from Office 365 to an On-Premise Exchange 2013 Server.

Questions:
If we set the autodiscover record to point to the new Exchange 2013 Server will the user's Outlook profiles automatically reconfigure themselves?

Will the user's simply get the typical, "The Microsoft Exchange administrator has made a change that requires you quit and restart Outlook" or can we expect more helpdesk intervention?

Depends how you are doing the migration.
If you are setting up a hybrid deployment, then moving the mailbox, then Autodiscover should make the changes for you.
However if you are doing anything else then you will be looking at new profiles.
