I want develop against SharePoint 365
I currently have Visual Studio Express for Web 2012.
Can I do this in that? What do I need?

Get yourself a Developer subscription to O365;

Download that NAPA tools on the same page.

You do not develop binaries for deployment to O365 as we used to for on premises development. Binary components are deployed to your own or cloud infrastructure.
