Question about changing int url's to match ext url's for .local support ending

I have the doc for the steps needed for this but my question is, should this be done after hours or can it be done during the day and if it is done during the day, what and how would the users be affected?

It can be done during the day.
It has zero impact on the users. It is information delivered to them by Autodiscover or when they browse to the server.

Ok Simon one more questions if I may?
If there are 2 exchange servers like in a DAG, I guess these commands to change the virtual directories would have to be run on both servers?
Is that correct?

All servers in the same AD site should be set to the same URL in most cases.
If on two different AD sites then you wouldn't use the same URL in both locations, as Exchange is AD site aware.
