Depends upon the Client PC's usage.
If the client PC's are just going to connect to Database Engine alone and perform basic development activities, then you can just use SQL Server 2008 R2 SSMS Express edition component (SQLManagementStudio_x64_ENU.exe) from
http://www.microsoft.com/en-us/download/details.aspx?id=30438

If they require access to SSRS, SSIS and SSAS along with advanced Management options like Profiler and DTA, then you would need to install SSMS from your SQL Server 2008 R2 Standard installer. Since you have 50 CAL licenses, you can officially install it in 50 PC's.

>> SQL server 2008 r2 standard  and 50 cal license

As you mentioned that you have a single SQL Server 2008 R2 Standard license, you are authorized to install SQL Server on a single Server.
And in addition to that you can install the Client tools on 50 Clients(machines) from where you can connect and work on the Server machine installed. So, you require CAL to install Client tools from the SQL Server installation media to connect and work on the Server.
