This should not be by design. I would attempt to create a new resource mailbox and then test the AutoAccept again. If this works it might be corruption with the mailbox itself. At this point what you could do is export the Resources Mailbox Calendar items and import them into the new Resource Mailbox.

As for removing the Deleted Items Folder you can use the following powershell command to accomplish this.
Give youself full access to the resource mailbox and the login to it using OWA delete the mail in the delete Items folder and test again.

Thank you Will.

The deleted items I now believe are not a factor, considering this Prohibit/Send at 5 GB.

Yes, other new room mailboxes work perfectly, accepting the room and sending notification. So I also believe that iit must be in fact corruption with the mailbox. But is there a way to definitively confirm that aside from locating an item in that mailbox that is "unopenable"? MFCMAPI is  cool but I would still need to know which items to look for in order to use MFCMAPI, in which case I could delete them. Do you concur?

Based on the issue you are having "Auto Accept not functioning" this is a core mechanism within a mailbox. It should simply work without any issues, if it is enabled. So based on that a lone you can determine that it is the mailbox as a whole and not just an items within the mailbox.

If you can create new Resource Mailboxes and everything works fine then there is something specifically wrong with this mailbox. There is no real way to determine this if it is not logged anywhere.
