Do you have Edge Transport server in place?
Please explain how many servers you have. E.g. Mailbox, CAS and Edge Transport.
Presently there is no scope to check Anti Spam agents in EAC, you will have to check and configure via Powershell only.

Thanks for responding and pretty new in Exchange and we have two exchange server.

Please open Exchange Powershell and run below command to see how many servers and if possible share those details here with changing host name
Get-ExchangeServer | Format-List
Else you can see Servers tab in left side of EAC.

we had four server roles for Client access as CAS001 to CAS002, CAS004 and CAS010 (SERVER ROLES as Client access)
Three Mail box server roles , MBX002,MBX004 and MBX010.

So it means you don't have Edge transport server.
See there are two ways:
Option:1
You can enable Anti-Spam on Mailbox server but it will be working with risk for direct attack.
Follow this article to enable Anti-Spam on Mailbox server.
Option 2:
Please install Edge transport server in DMZ network which will face all e-mails coming from internet. Edge transport server is already configured with Anti Spam features, you just need to change condition as per your requirement e.g. whitelisting e-mail address, blocking e-mail address or blackilisting IP address.
In case you don't want to invest on hardware/software so you can hire a cloud anti-spam solution like Exchange Online protection, message labs or postini.. Lot of solution are available.

For your information that we are using the Exchange Admin Center 2013.
Question how to check the spam gateway in Exchange Admin Center?
