Have some spyware on a computer that I can't seem to get rid of or figure out.  I have run malware bytes, spybot, super antispyware, but none have worked.  Take a look at the screenshot.

Short of a system restore or a wipe and fresh install, I would try RKill and Combofix.

Will Rkill fix be permanent or will I have to run it each time I boot?

You run rkill and then combofix or malwarebytes afterwards, you don't restart until the malware has been quarantined/removed.
