I wear a lot of hats...

"The solutions and answers provided on Experts Exchange have been extremely helpful to me over the last few years. I wear a lot of hats - Developer, Database Administrator, Help Desk, etc., so I know a lot of things but not a lot about one thing. Experts Exchange gives me answers from people who do know a lot about one thing, in a easy to use platform." -Todd S.

No, this record needs to be configured when you move over to Exchange Online and you no longer have any mailboxes on-prem. For Hybrid, Autodiscover should point to your local server.

our major consideration for moving  to office 365 , critical mail users  high availability  .

if on premises server is down we can continue to access mail from  office 365 for critical mail users

you can't have 2 records and a cname as the results of a dns lookup will be unpredictable.
a autodiscover 10.1.2.3
CNAME Autodiscover autodiscover.outlook.com

Don't point autodiscover to O365 in Hybrid Exchange scenario
Just keep it AS IS - onpremise
Your hybrid setup co-existence can't work with autodiscover pointing to O365
As suggested earlier by others, complete all mailbox movement in cloud and then switch autodiscover
You may switch MX if wanted to

The concept you are exploring works well for migrating email platforms other than Exchange to O365

Also if you want to authenticate O365 users with email and password only, configure directory sync with default option, Objectguid with userprincipalname under user matching
Make sure that users UPN (Ex:user1@domain.com) should match their primary smtp address
Now Use Dirsync password sync feature OR you can use Adfs to get SSO
