I need real life hyper v 2012 r2 disaster recovery scenarios with best practices in regards of business continuity and least down time.

Have you already setup 2nd hyperV host or cluster ?

Yes, with replication and replica broker.

A known good backup.

Hyper-V Replica does _not_ replace a known good backup setup.

That being said Host 1 --> Host 2 --> Host 3 in a Data Centre.

Back up Host 1, test that backup by fully restoring the entire set to another server, then you're good to go.
