Enable Smooth edges of screen fonts for Remote Desktop users

I understand the Font smoothing setting is an option within the Remote Desktop connection software however, is it possible to force this "on" on our RDS 2012 R2 server?

Not sure what you're asking.    You have to set this on the client computer by going to the "Options - Experience" tab for RDC and choosing "font smoothing".     Once you've set it, it should be persistent as long as you log in with the same user credentials as you were using when you set it.

I am aware the user can select the option to use font smoothing in RDC however I would like to ensure it is switched on regardless of what option the user supplies

Is this possible?

As far as I know you can't force this unless you're logged in and use the normal process [i.e. Options - Experience].
