we have hundreds of emails we need to add in block sender list in proofpoint(gui)

is there any script we can run

Put all the email addresses in Microsoft word in a list one underneath each other.   To copy and past them in to proofpoint they will need a semicolon.   To that in word you will have to run a find and replace. In Microsoft word with your list of email addresses. Find>advanced find click Special, click Paragraph mark. Now click the replace tab.  put in the ; then press the space bar one time.  
Then click replace all.  You will now have your email addresses formatted with a semicolon in between. Copy and paste this now in to your block senders list.

what you mentioned about word is perfect but how should I get 100 email address from proofpoint addressed to that user, how to filter that, can I export or do I need to add manually in word

after you have all the email addresses separated by semicolons can you not highlight all, copy and paste them in to proofpoint blocklist?
