how do i publish an application,

First of all, you do not need to publish a web app into your local machine.
Vs2010 will create that for you every time you build/rebuild the solution, this is important because you would like to debug your application.

My suggestion will be to open your app and rebuild the solution. Then you can press F5 or run without debugging to check that your app is operational. To be on the safe side, open IIS manager and have a look that VS has created your website.

After that you can use your site with out using VS by just calling the correct URL.

NOte: If you still cannot make it work, first of all create a simple hello world app and try steps above. I it works then your issue could be errors in your web.config that you will need to attach to the question.

An example from MSDN. Do not forget to rebuild solution before that. Just remember that running your website will publish it automatically to your local PC. Just make sure that IIS is properly installed in your PC. (there are plenty of tutorials in you tube/google depending on OS)
Note: IF more help needed please post OS version of your PC.
