If you have access vlan401 between 7250 and 6509, I am not sure if PC2 can ping PC1.

I have never thought of your second design. That is very interesting. Thanks
Kindly confirm if the links connecting the L3 switches are trunk links.
There are default gateways defined on the L3 on the right hand side for vlans 401 and 404.
