I want to install win 2012 r2 datacenter and hyper -v on dl585 with 4 16 core processors

I have a valid license number for 2012 r2 datacenter.

If for example I setup hyper-v and I want to create 50 windows 10 desktops will I also need licenses for those as well?

What about cals for the clients that will be connecting to these desktops?

You mean remote desktop services right? Yes you need RDS calls.
You mean 50 vm's? Yes you need 50 licenses

As i remember correctly, on a 4 cpu machine you need two datacenter licenses.

Datacenter allows for 2 CPUs so you need 2 licenses...
As for the VMs you can have unlimited VMs but only server editions...

Yes... You are correct the host will need 2-licenses..

Do you know if hyper-v will run persistent and non-pertsisent desktops like vmware's vdi?

It seems that Hyper-V supports both although i don't have 1st hand knowledge ...a bit of googling revealed a lot of articles on this subject...

Yes hyper-v supports non persistent desktops in a VDI.
Don't forget about your CAL's.

Server 2012 R2 DC is Proc + CAL on the licensing. The Win desktop machines will require its own license.
