VM sizing

I have a cluster of 3 esxi hosts, each host has 2 x 8-core CPU and 256GB RAM; datastore size 6TB.
Is it realistic to run a MS SQL server VM with 16 core, 128GB RAM, and 3TB disk space on this cluster?

Yes, depends on the other free resources you have available.

and do you really require 16 sockets for your VM.

well, you could also think of a separate physical deployment here.
But vSphere is still an option, sure, also there might not be the need to configure 16 vcpu for this VM..

Virtualization in your case :
- ease of use (vmotion and all vmware potential)
- easier protection (backup/restore, replication)

- it will already take a big part of your vmware cluster ressources
- less performance than without virtualization
