VM fileserver and VM storage space (>2TB?)

Hello every one,

Sorry for asking these question in maybe not the best English, as it is not my native language. I`m also not a VMware professional but I hope you understand my questions. I have two questions about current best practise virtual fileserver. We are currently using vCenter Server v 6.0.0.
We have a fileserver which has hdd`s (volumes) mounted by iSCSI. These are stored on a dell EqualLogic. In the last article I read they said if you can use VMFS -5 of VM virtual hdd, it is better to use a virtual HDD for fileserver files than mounting hdd by iSCSI. Is this the actual best practise?
Also if these volumes store a big amount of files (500gb-1500gb) does this work well with VDP or is this a problem?

Second question is if there are any (more) restriction about volume size in vSphere where you store your VMs at (I mean not the hdd in vms). I remember with vSphere v. 3.x there was a problem when using more than 2TB for your VM Data Store. But with 6.0.0 we shouldn’t have any problems using a big Volume to store all VMs as I understand https://kb.vmware.com/selfservice/microsites/search.do?language=en_US&cmd=displayKC&externalId=2058287 correctly, right? But this seems to cover only HDDs IN VMs where I mean the Store where you put your VMs in…
Also I have heard that a mega volume for all VMs is not really the best in case of possible IO problems. Is this still the case?
