I have a software that uses the graphic card GPU's .

I have the Nvidia Quadro M6000 connected to the server. I need to run 5 sessions of this program.

I was told VDI is the way to go.
VDI is literally a full Windows client VM per user. It isn't multiple sessions like RDS.   And the licensing for such a setup is rather expensive.  Windows with SA,  or VDA,  or managing multiple retail licenses with keys,  and hardware beefy enough to run that many VMs. Sounds like that wasn't quite what you were expecting.

I want one machine (server) to host the hardware , and allow 5-6 regular computers to run the software that usages the graphic card.

I just want to find the best way to do it. at this stage cost isn't an issue.

But, I have a server that I need to work with what I have.
Which is It? Is cost not an issue? Or do you need to work with what you have?  Those are contradictory statements.

I'll be blunt. VDI is an advanced management scenario and requires a very skilled I.T. team to handle properly. It doesn't sound like it is a good path given your questions.

So what do you suggest.

Describing the scenario im at .

I have a server with the m6000 graphic card. and need to run 5-6 sessions , with the program. ?
here are some walkthrough guides

https://docs.microsoft.com/en-us/windows-server/remote/remote-desktop-services/rds-remotefx-vgpu

http://www.vdicloud.nl/2010/11/27/step-by-step-installation-guide-installing-a-rd-session-host-with-remotefx-part-3/

But you know my overall thoughts and recommendations, as to what you are attempting outlined in the other question

If this was a team of AutoCAD users, with little need for AutoCAD, and small 3D CAD files, fine, but 3D App Developed and needing CUDA cores!

Workstations are the way to go...
Add-VMRemoteFx3dVideoAdapter -VMName VirtualMachineName

Also ensure you are using the latest drivers on the host machine

I dont' understand what do I need to do .

I have the server with this graphic card. I want to allow users to use the program . what should I do?

Also regarding licences, if I have 100 users but want to allow only 10 concurrent session. how can I achieve that?

Its worth noting that the guide is for the release candidate of 2008 R2, was never a supported method, and will often bluescreen 2012-era drivers and newer.  RemoteFX vGPUs are a VDI technology and not meant for RDSH.  The multi-user nature of RDSH just causes issues with trying to use it.

https://docs.microsoft.com/en-us/windows-server/remote/remote-desktop-services/rds-remotefx-vgpu

Not that even in 2016, the only time a vGPU is supported with the session host role is if it is configured as a "personal session" which is effectively VDI using a server OS instead of a client OS.
OR a more clear-cut answer:

https://docs.microsoft.com/en-us/windows-server/remote/remote-desktop-services/rds-supported-config

Note the first column for vGPUs.  2012 R2 RDSH and 2016 RDSH specifically got their own rows since RDSH effectively kills the vGPU driver.
