Hi,
what does it mean "enable centralized mail transport"
If I enable it during hybrid wizard ?
I need to configure hybrid but all mailboxes shoud stay on-premisse and all mail traffic should happen via on premisse exchange.

If you need all mail traffic to be routed to the on-premises Exchange, including that for cloud mailboxes, that's the option you should configure. Read more here: https://technet.microsoft.com/en-us/library/jj659050(v=exchg.150).aspx

If all mailboxes are staying on-prem, then don't select "Enable Centralized Mail Transport.", that way your on-prem Exchange server will route mail for your org O365 will use the dedicated send connector for internal mail flow between O365 and on-prem.

If you select that option, whenever your cloud mailbox users send emails to internet users, the mail 1st forwarded / relayed to onpremise Exchange server via O365 to onpremise connector created by hybrid wizard and then its onpremise exchange server responsibility to send out that email to internet
This setting is done by few organizations as part of there compliance / security team requirement / enforcement that all external emails should pass through onpremise outbound email gateway
Otherwise O365 mailbox users external email will directly reach to internet via O365 smtp outbound gateway (EOP) and onpremise exchange emails to internet would flow directly to internet via onpremise outbound smtp gateway

That simply means if you have hybrid setup all the emails will routed to on premise exchange transport system and than goes to respective recipient.

Organization usually do this for compliance and for admin purpose this is a good option however this brings more configuration part where you have to have proper SSl certificate and make sure TLS 1.2 is enabled on on prem servers as well
