O365 hybrid and autodiscover


I´ve an exchange 2010 hybrid scenario to O365.

Autodiscover points to  On-Premise and mails accounts are migrated 10-20/week.

When should be changed from On_premise to O365 the autodiscover registry?

In general, in Hybrid it's best to leave it pointing to on-premises. Only after you've migrated all mailboxes and have no plans for creating new on-premises mailboxes (or migrating them back from O365) you can change it to point to O365.

Right, then for migrated accounts to O365 and Autodiscover poiting to On-Prem, after moving them, users should restart outlook and everything would be ok poiting to cloud?

They *might* be prompted for restart, or for credentials depending on few factors. But in a nutshell, yes, that's what should happen end everything should work OK.
Issues you may encounter and which have to be accounted for in case of a large migration:
OST corruption
Profile corruption
In both cases you need to recreate outlook profile.
Microsoft says percentage to be below 2% of migrated mailboxes.
I have experience of an higher percentage, about 10%
