AppLocker question does AppLocker have any affect on applications already installed?  When applying updates for example we use WireShark when moving from 2.6.3 to 2.6.4 does anything need to be added to AppLocker to allow the update?  Can AppLocker be managed from Active Directory?

Yes, if it is blocked it won't run.

When applying updates for example we use WireShark when moving from 2.6.3 to 2.6.4 does anything need to be added to AppLocker to allow the update?
Depends on how you configured AppLocker. If you used a reference computer to whitelist EXEs and WireShark wasn't there, yes.

Can AppLocker be managed from Active Directory?
Yes, you can manage it via a GPO

Okay, so does there need to be a "WhiteList" entry for each update for WireShark, or just a blank exception for WireShark?

It is up to you but to continue to get updates you need an application one

you could also white-list the publisher
