Full Install of Microsoft Exchange 2016 CU12: Configuring ETL traces before fast service is installed.
Fresh install of Server 2012 R2, now trying to install Microsoft Exchange 2016.
Step 6 of 13: Mailbox role: Transport service

We have disabled Firewall Services and it is set to Manual.

We have stopped the windows firewall service and disabled antivirus.
Did you reran the setup and r u facing same issue.
What is the version of Exchange you are using?
Then, take steps below to check the version of .net:
1. On the Start menu, choose Run.
2. In the Open box, enter regedit.exe.
3. You must have administrative credentials to run regedit.exe.
4. In the Registry Editor, open the following subkey: HKEY_LOCAL_MACHINE\SOFTWAR

Make sure the version of .net is at least 4.6.2, other wise you should update .net first then install Exchange 2016 CU9.
Then, take steps below to check the version of .net:
