launch RDP with a URL
Have a client that has a virtual assistant company, as a call comes In that client profile comes up in his system, at some clients he needs to connect to a computer at the client's since he needs to use a locally installed application, I would recommend him to you RDP but since the system he uses only accepts a URL, is my question is there is a way to launch RDP with a URL?

connectwise will only give me a url to the portal and from there I would select the client to connect, I would prefer if I can have a url that goes directly to the client PC
The only way I can think of that you can use RDP would be to
VPN Method:
Connect to a VPN and then RDP to said machine
Port Redirection Method (Very unsecure)
Setup a unique port on the firewall and redirect all traffic on WAN IP and Port # to Host:3389

VPN will not work since he is connecting to many clients and thy be on the same network

Most RDP applications aren't going to allow this by default. You could also use Team Viewer Enterprise, but that is a little more expensive.
