Will this memory work with my Dell Optiplex 7040

Here is memory on sale at Newegg for $85.

G.SKILL Aegis 32GB (2 x 16GB) 288-Pin DDR4 SDRAM DDR4 2666 (PC4 21300) Desktop Memory Model F4-2666C19D-32GIS
DDR4 2666 (PC4 21300)
Timing 19-19-19-43
CAS Latency 19
Voltage 1.2V

Here is compatible memory according to Crucial memory scanner:
DDR4 PC4-19200 • CL=17 • SINGLE RANKED • X8 BASED • UNBUFFERED • NON-ECC • DDR4-2400 • 1.2V •

I currently have 4 sticks of 4GB memory. I'd take out two and substitute the 16GB sticks. Will this work?

You need to update your BIOS. You can generally buy either one but you will only go as fast as your motherboard will support.

But my suggestion is keep similar type as recommended by Crucial site and DELL
