What are the differences between AD Distribution List & Exchange Distribution Group?

Does a GPO apply to AD distribution groups too?

I know NTFS permissions do not apply.

It should not apply if its just Distribution group as it will not have the SID and nay permission or security related settings can be applied only if it has SID.

Distribution group in Exchange: just a place to distribute emails through all members of the group

Distribution group in AD: A group of AD users. This one can be configured permissions so that ones can access shared files/folders whilst others cannot

There is no such thing as Exchange Distribution groups. What you are referring to as Exchange Distribution groups is still an AD group, even though you used the exchange console to create it. There 2 Distribution groups, and they are security group and distribution group. The difference is the security group can have permissions assigned to it and also have GPOs apply. The normal distribution group can't have permissions assigned and it will only be used to accept emails that is distributed to the members of that group but no AD permissions or GPOs are assigned to it, while the security group can get AD permissions, and GPO applied and it can also accept mail for the members of that group.

if that’s the case, should I be creating AD distribution groups? Or the distribution groups in Exchange will suffice?

If the distribution groups are for email purposes, just create distribution groups in Exchange admin center or O365 admin center for simplicity.
