Troubleshooting Question

Can't get Autodiscover working properly on a Exchange 2010 Server

Have tried changing DNS but all to no avail as still can't get Autodiscover working, have also tried with using a VPN to the Server but still doesn't't work, hopefully something easy I'm missing.
