We've got a virtual (hyper-v) exchange 2016 server.
The host hyper-v has 2 vhdx files on it, serving as the 2 hard-drives of the exchange machine; one is for the exchange's OS & the other contains the DB only.
These 2 vhdx files are sitting on the same physical hard-drive back at the hyper-v host server, which is starting to fill up.
Can I simply move the Mail-DB.vhdx file (the Exchange's hard-drive on which we only keep the Exchange DB) from one drive to the other on the host machine, and simply point to the virtual hard disk's new location?
Is there anything I'm missing? any specific guidelines? something to keep in mind or to be aware of?

in general yes...Moving vhdx files on the Hyper-V host doesn't really affect exchange. Exchange reflects on the drive letter. The drive letter has to be the same.
From the general procedure...
- Shutdown Exchange...
- Detach the drive
- Move (or copy) the drive
- Reattach the drive...
- Restart exchange...
- Check if the drives appear as expected.
What you may have in mind...
A new physical drive can have different permission. As far as you already store vhdx files on the new location, they should be fine. If you now use a new drive the first time, you first have to prepare the new drive location so the containing folder has the correct permissions.
The permission for the Hyper-V machine itself is set, when the disc is attached.
